تاثیر افزودن نانوذره سلنیوم بر بیان ژن آنزیم استئارویل کو‌آنزیم‌آ ‌اشباع‏زدا در بافت چربی بره‏های نر قبل از بلوغ جنسی

چکیده

سلنیوم در ساختمان سلنوپروتئین و هورمون­ها نقش حیاتی دارد. فعالیت آنزیم استئارویل کو­آنزیم­آ اشباع‏زدا منجر به شکل­گیری اسیدچرب غیراشباع تک­رشته­ای از اسیدچرب اشباع می­شود. مطالعه حاضر جهت بررسی اثر تجویز خوراکی نانوذرات سلنیوم بر میزان نسخه­برداری ژن آنزیم استئارویل کو­آنزیم­آ اشباع‏زدا بافت چربی دنبه بره­های نر قبل از بلوغ جنسی انجام شد. بدین­منظور چهارده رأس بره نر تازه از شیر گرفته انتخاب شده و در گروه‏های تیمار 1 و 2 (هر یک 5 رأس) و شاهد (4 رأس) تقسیم شدند. نانوذره سلنیوم طی 20 روز متوالی با دوز 05/0 و 1/0 میلی­گرم به ازاء هر کیلوگرم وزن بدن به ترتیب به گروه­های تیمار 1 و 2 و آب مقطر با حجم مساوی به گروه شاهد خورانده ­شد. نمونه­برداری از بافت چربی دنبه در روزهای صفر (قبل از خوراندن نانوذره)، 20 و 30 صورت گرفت و میزان بیان ژن آنزیم استئارویل کو­آنزیم­آ اشباع‏زدا به­روش RT-PCR تعیین شده و داده‏های حاصله با بهره‏گیری از آزمون آنالیز واریانس یک‏طرفه در سطح 05/0p< مورد تحلیل قرار گرفت. نتایج نشان داد که قبل از تجویز مقادیر مشخص­شده نانوذرات سلنیوم و آب­ مقطر، اختلاف آماری معنی­داری بین گروه­ها در میزان نسخه­برداری ژن آنزیم استئارویل کو‌آنزیم­آ اشباع‏زدا وجود نداشت، اما در روز 20 آزمایش بین گروه­های تیمار 1 و 2 اختلاف معنی­داری شکل گرفت (05/0p<). نتیجه این‏که تجویز نانوذرات سلنیوم با دوز 1/0 میلی‏گرم برای هر کیلوگرم وزن بدن به‏مدت 20 روز متوالی موجب افزایش بیان ژن آنزیم استئارویل کو­آنزیم­آ اشباع‏زدا و بهبود کیفیت اسیدهای­چرب موجود در بافت دنبه می‏شود. به عبارت دیگر برای اولین بار نقش تأثیرگذار نانوذره سلنیوم بر شکل‏گیری اسیدچرب غیراشباع توضیح داده شد.

Selenium play a crucial role in the formation of selenoprotein, hormones and intrinsic factors. The stearoyl-CoA desaturase enzyme (SCD) is present in many tissues of the body and its activity leads to the formation of a single-stranded fatty acid from saturated fatty acids. Present study was carried out on fat tail weaning male lambs to evaluate the effect of oral selenium nanoparticles administration on transcription rate of stearoyl-CoA desaturase enzyme. For this purpose, 14 male lambs were selected at weaning time and divided into three groups: treatment 1 (5 heads), treatment 2 (5 heads) and control (4 heads). Selenium nanoparticles were administered for 20 consecutive days at a dose of 0.05 and 0.1 mg/kg B.W. to treatment groups 1 and 2, respectively and distilled water to the control ones. Tail fat tissue samples were collected at days 0, 20 and 30 via core biopsy needle gauge 14.
